Obtaining of Plantlets through Immature Embryo Culture of Lilies


Abstract
This experiment was conducted to improve fertilization rate, to investigate cultural conditions of immature embryo, and to observe process of embryo development in vivo in Lilium. The fertilization rate could be increased by cut-style pollination. It was possible to culture 4th week immature embryos of 0.14§® in length. Optimal age of embryo for immature embryo culture of lily plants was 6th week after pollination. For culture conditions of immature embryo, MS medium with 3% sucrose (L. longi,florum cv. Georgia), 0.7% agar (L. longiflorum cv. Hinomoto), NAA 0.01ppm (L. longiJforum cv. Hinomoto), and l6hrs day length (Enchantment X L. concolor) were recognized as the most suitible. For transplanting obtained plantlets into soil, cold (4¡­5¡É) or submerging (45¡É) treatment was not needed. Microscopic observations indicated that embryo was developed into globular (1th week after pollination), semilinear (4th week after pollination), and linear types (6th week after pollination) and aborted (5th week after pollination) occasionally.
